Hugh and Leifson의 OF 배양은 아래의 탄수화물발효 배지와는 다르다.
peptone의 농도는 1%~0.2%로 저하시킨다.

agar의 농도는 반고체상태로 만들면서 1.5%~0.3%로 감소시킨다.
단백질/탄수화물의 비율이 낮을수록 아마 산화적 대사로부터 형성된 약한 산의 적은 양을 중화시키는 알칼리 amine의 형성이 감소한다. 하지만, glucose의 산화적 분해로 형성된 산들은 매우 약하고, Hugh and Leifson의 더 민감한 산화-발효 배양 (OF medium)은 발견이 요구되어진다.
